| Abstract: Fluorescence has proven to be a versatile tool for a wide range of applications in physical and life sciences. It is a powerful technique for studying immediate molecular environment, inter and intramolecular interactions. In this paper is presented an overview of how to set-up luminescence assay, mainly regarding to the excitation, emission and time drive recording spectra. Additionally, we discuss how to troubleshoot a luminescence assay, how to identify the various scatter artefacts, and the influence of polymer concentration and the solvent on fluorescence spectra. The practical experience is demonstrated by our concrete results of rotaxane florescence recording spectra. |
